| Spanish abstract | Un método para el diagnóstico de la fiebre tifoidea es la serotipificación que se basa en el principio de aglutinación a partir de los antígenos flagelar H y somático o con sus anticuerpos correspondientes. Por lo tanto, el objetivo de este trabajo de tesis fue desarrollar un reactivo de diagnóstico para dicha enfermedad por seroaglutinación. Para tal fin, la cepa Salmonella typhi ATCC 6539 se propagó en Agar Sulfito de Bismuto, comprobando su pureza por morfología colonial y microscópica, y por pruebas bioquímicas. Posteriormente, se produjeron los antígenos H en caldo flagelar y caldo formolado con fucsina, y el antígeno O en caldo soya tripticasa y caldo fenolado con rojo Congo. Los antígenos se evaluaron con sueros positivos de conejo inmunizado y de pacientes de un nosocomio del Estado de México, observando resultados positivos por aglutinación significativa |
| English abstract | A method for the typhoid diagnosis is serotyping which is based on the principle of agglutination from the flagellar H and somatic O antigens and their corresponding antibodies. Therefore, the aim of this thesis was to develop a diagnostic reagent for this disease by seroagglutination. For this, the strain of Salmonella typhi ATCC 6539 was propagated in Bismuth Sulfite Agar, verifying its purity by colonial and microscopic morphology and biochemical tests. Subsequently, the H antigen was produced in flagellar and formolade broths with fuchsin and the O antigen was produced in trypticase soy and phenolated broths with Congo red. The antigens were evaluated with positive sera from immunized rabbits and patients from a hospital in State of Mexico, they produced positive results by significant agglutination |
