Resumen en inglés | Rosmarinus officinallis Linn. (rosemary), of the family Lamiaceae, is rich in aromatic essential oils and exploited due its pharmaceutical, aromatic and culinary properties. Seeds and nodal segments of rosemary were submitted to different conditions of surface sterilization and placed on Murashige & Skoog (MS) culture medium, with the aims of investigate the germination viability of seeds and determinate the best sterilization conditions of meristematic tissues for the in vitro production of plants. Two concentrations of sodium hypochlorite (NaClO) solution (0.5% and 1%) were tested for 10 minutes for seed sterilization, as well as the control (0% de NaClO), with further culture on MS medium. For the sterilization of nodal segments, two concentrations of NaClO (1 and 1.5%) were tested for 5, 10, 15, 20, 25 and 30 minutes. Further, 1% NaClO was tested (5, 10 and 15 minutes), with previous immersion in 70% ethylic alcohol. The nodal segments were individually cultivated in test tubes containing MS medium supplemented with 6-benzylaminopurine (BAP) and a- naphtaleneacetic acid (NAA). Contamination with fungi and bacteria, surviving of the explants as well as shoot formation were evaluated. On seeds, the minor contamination was observed applying 1% NaClO. Germination rate was very low (0.67%). The nodal segments submitted to sterilization for 1.0% NaClO with previous immersion in 70% ethylic alcohol for 10 and 15 minutes presented minor contamination rates (45 and 30%, respectively) and major surviving and shoot formation percentages (100 and 78.5%, respectively) |
