Resumen en inglés | The molecular mechanisms that control protein functions have been a field of extensive research and new knowledge. It is known that there are control mechanisms for the oxyreduction of specific amino acids in protein-sensitive sites such modification. The introduction of disulfide bonds, inter- and/or intrachain, is between these modifications may be responsible for the functional control of various proteins, e.g. insulin. Oxyreductases reactions are catalyzed mainly by family members PDI, are crucial to the performance of these reactions of oxyreduction. The QSOX, quiescin sulfhydryl oxidase, performs the insertion of disulfide bonds in nascent and mature proteins through the reduction of molecular oxygen to hydrogen peroxide. The biological role of this enzyme remains unclear, however the literature suggests a role in intra- and extracellular environments, participating, for example, the folding of extracellular matrix molecules, and possibly by activating signaling pathways responsible for cell survival. Resumen en portugués | Os mecanismos moleculares que controlam as funções proteicas têm sido um campo de ampla pesquisa e novos conhecimentos. Sabe-se já que há mecanismos de controle pela oxirredução de aminoácidos específicos em sítios proteicos sensíveis a tal modificação. A inserção de ligações dissulfeto, inter e/ou intracadeia, está entre essas modificações que podem ser responsáveis pelo controle funcional de diversas proteínas, como a insulina. As reações oxirredutases são catalisadas, principalmente, por membros da família PDI. A QSOX, uma quiescina/sulfidril oxidase, realiza a inserção de pontes dissulfeto em proteínas maduras e nascentes, por meio da redução do oxigênio molecular a peróxido de hidrogênio. O papel biológico dessa enzima permanece obscuro, contudo a literatura propõe a atuação em ambientes intra e extracelular, participando, por exemplo, do dobramento de moléculas da matriz extracelular, e possivelmente ativando vias de sinalização responsáveis pela sobrevivência celular. Nosso grupo de pesquisa tem como intuito desvendar alguns possíveis substratos e mecanismos biológicos dessa proteína in vitro |
