Spanish abstract | La conservación de hongos en las colecciones frecuentemente induce cambios atípicos de sus ejemplares. En este trabajo se evaluaron diversos medios de cultivo para recuperar las características morfológicas de los dermatofitos conservados en promedio 10 años y conocer su patrón de sensibilidad a nueve antifúngicos. Se estudiaron 54 aislamientos, de ellos 15 primarios aislados recientemente y 39 de colección, incluyendo la cepa ATCC 9533 de Trichophyton mentagrophytes. La mayoría de los aislamientos corresponde a T. mentagrophytes, T. rubrum and T. tonsurans. Todos los dermatofitos se crecieron en agar dextrosa Sabouraud y después se inocularon en cinco medios diferentes para detectar en cuál de ellos se recuperaba la morfología típica de las especies. Para determinar la sensibilidad antifúngica se aplicó el método de microdilución en caldo, de acuerdo al documento M38-A2 del Clinical and Laboratory Standards Institute. La mayoría de los dermatofitos de colección mostraron características coloniales atípicas al inicio del estudio. Después de ser inoculados en los diferentes medios, el agar Gorodkowa modificado, el agar avena sales y el agar tierra-pelos indujeron la producción de numerosos conidios. En este último, además se formaron órganos perforantes. El 85 % de los aislados fueron inhibidos por la terbinafina (0.0008 y 0.03 μg/mL); la caspofungina y anidulafungina mostraron baja o nula actividad |
English abstract | The preservation of fungi in collections frequently induces atypical changes of the specimens. In this work, several culture media were evaluated in order to recover the morphological characteristics of dermatophytes preserved in average for 10 years and to determine their susceptibility pattern to nine antifungal drugs. Fifty-four isolates were studied, from them 15 primary isolates and 39 from collection, including the ATCC 9533 Trichophyton mentagrophytes strain. The majority of the isolates correspond to T. mentagrophytes, T. rubrum, and T. tonsurans. All dermatophytes were firstly reactivated growing them in Sabouraud dextrose agar and then cultured on five different media containing diverse ingredients to evaluate the species morphology. For the susceptibility studies, the broth microdilution method was used, according to instructions of the document M38-A2 (Clinical and Laboratory Standards Institute). Most of collection' dermatophytes initially showed colonies with atypical morphology. After their growth on different media, modified Gorodkowa agar, oat-salts agar, and soil-hair agar induced numerous conidia production. In the last one, also perforating organs were formed. Terbinafine inhibited 85 % of dermatophyte isolates (0.0008 and 0.03 μg/mL); caspofungin and anidulafungin showed low or null activity |
