Spanish abstract | Las relaciones genéticas entre colonias de anidación de caguama en el suroeste de Cuba (Península de Guanahacabibes, Cayería San Felipe, sur de la Isla de la Juventud y Cayo Largo del Sur), se determinaron usando secuencias de la región de control del mtDNA (375 pb del extremo 5’). Estos datos fueron analizados en un contexto regional para estimar la contribución de cada colonia a la estructura genética de la metapoblación del Gran Caribe y la costa atlántica de USA, empleando tablas de contingencia de χ 2 y Ф ST. Las incongruencias entre algunos de los resultados de estas pruebas fueron resueltas probando hipótesis de asociación genética y geográfica a través de AMOVAs. Dentro de las colonias de anidación cubanas, fueron encontrados haplotipos pertenecientes a dos linajes, i. e. norte y sur del Atlántico [A]; Gran Caribe – Mar Mediterráneo [B]. Un haplotipo perteneciente a este último (CC-A2), estuvo en alta frecuencia respecto a los demás (CC-A1, 8, 10, 12, 14). El haplotipo CC-A12, previamente reportado para áreas de forrajeo, resultó endémico de la Cayería San Felipe. No se encontraron diferencias significativas entre las colonias cubanas pero se encontró estructura genética cuando éstas fuer on comparadas con las demás colonias de la región. La población de anidación cubana fue considerada como un a unidad demográfica independiente al igual que las colonias de anidación del oeste y este del sur de la Florida. Por consiguiente, la población de anidación cubana constituye un demo de una metapoblación, que debe ser manejada en un contexto regional |
English abstract | Using ca. 375 bp sequences from the 5’-end of the mitoch ondrial DNA control region we elucidated the genetic relationships among four loggerhead rookeries in sout hwestern Cuba (Guanahacabi bes Peninsula, San Felipe Cays, south of Isla de la Juventud and Cayo Largo del Sur), and analyzed these data within a regional context to understand the contribution of each to the genetic structure of the Greater Caribbean metapopulation. Contingency chi-square tables and Ф ST tests we applied to all pairwise comparisons of Cuban and previously analyzed rookeries for the species in the Atlantic basin. Incongruities between some of the results from these tests we resolved by testing hypoth eses of genetic versus geographical association through AMOVAs. Within the Cuban rookeries, haplotypes belong to two different lineages, i.e. North and So uth Atlantic [A], and the Greater Caribbean – Sea Mediterranean [B] lineages. On e haplotype (CC-A2) belongin g to the latter, was in higher frequency respect to the others (CC-A1, 8, 10, 12, 14). One haplotype (CC-A12), previously only reported from foraging areas, was found to be endemic for the San Felipe rookery. No significant differentiation was found between any pair of Cuban rookeries, when the set of Cuban rookeries was compared to the remaining rookeries in the analyzed region, some genetic structuring was revealed. The Cuban population was considered as an independent demographic unit the same as the western and eastern rookeries of the south of the Florida. Cuban nesting population constitutes a deme of a metapopulation, which should be managed in a regional context |
