English abstract | The present research aimed to evaluate the effect of different light spectra on seed germination and initial in vitro development of Phyllanthus tenellus Roxb. Furthermore, N concentration was evaluated in plant development. Seeds were sterilized and inoculated in solid MS medium. Seeds and plantlets were maintained under the following light treatments: white (control), blue, green, red and white plus UV-A. Seed germination was observed under light and darkness. Under red light, germination showed the greatest percentage (12%) after 12th week, considered very low. Germination rate also increased under red light. Rooting was favored by red light (80%) and white light (71.4%), and reduced under blue light (51%). As results from blue light exposure plantlets presented the highest number of leaves. MS medium reduced to half of NH4NO3 and KNO3 solution improved rooting and revealed the best conditions to in vitro establishment of P. tenellus cultures |
Portuguese abstract | Este trabalho teve como objetivo avaliar a germinação de sementes e o desenvolvimento inicial in vitro de Phyllanthus tenellus sob diferentes faixas do espectro luminoso, bem como o uso do meio MS com redução da concentração de nitrogênio no desenvolvimento das plântulas. As sementes foram desinfestadas e inoculadas em meio MS sólido. As sementes e as plantas foram mantidas sob os seguintes tratamentos de luz: branca (controle), azul, verde, vermelha e branca acrescida de UV-A. As sementes germinaram tanto sob luz, quanto no escuro. A máxima porcentagem de germinação foi obtida na 12ª semana, sob luz vermelha (12%), considerada muito baixa. A luz vermelha também aumentou a velocidade de germinação. A porcentagem de enraizamento foi maior sob luz vermelha (80%) e branca (71,4%), e reduzida sob luz azul (51%). A exposição à luz azul resultou em plântulas com maior número de folhas. A redução à metade das concentrações de NH4NO3 e KNO3 do meio MS aprimorou o enraizamento, adequando as melhores condições para o estabelecimento in vitro das culturas de P. tenellus |
