Resumen en español | Se ha considerado la posible función del polimorfismo (TTTA)n del gen CYP19, que contiene un número variable de repeticiones TTTA (7 a 13), en el cáncer de mama. Esta estructura interviene en la biosíntesis de los estrógenos y se ha reportado un incremento del riesgo de padecer cáncer mamario cuando las mujeres presentan > 10 repeticiones alélicas de (TTTA)n. Objetivo: Analizar la frecuencia de los diferentes genotipos del polimorfismo de repetición simple (TTTA) del gen CYP19 en mujeres sanas y pacientes con carcinoma mamario. Material y métodos: Se incluyó a 180 mujeres sanas y 70 con cáncer de mama. Se obtuvo el ADN de los leucocitos para la genotipificación. Las repeticiones de TTTA de CYP19 se determinaron por amplificación con PCR y los productos amplificados se corrieron en gel desnaturalizante de acrilamida. Resultados: Se incluyó a un total de 250 mujeres de 55 ± 12 años con IMC de 30 ± 7.1 kg/m2. Se reconoció una distribución diferente en las frecuencias de los alelos CYP19; en las mujeres sanas, los alelos más frecuentes fueron 6 (32.7%) y 7 (21.6%) y en las mujeres con cáncer las repeticiones alélicas más comunes fueron 6 (29%) y 10 (26%). Conclusión: Este estudio indica que el número de repeticiones >10 TTTA puede estar relacionado con un mayor incremento del riesgo de padecer cáncer mamario |
Resumen en inglés | It has been considered to be the possible role of the (TTTA) n polymorphism of the gene CYP19 in the cancer of breast. They contain a different number of TTTA repetitions (range 7-13) inside the region of codification. It is involved in the synthesis of the estrogens and an increase in the risk has been reported when the patients present >10 repetitions allelic of (TTTA)n. Objective: Analyze the frequency of the different genotypes of the polymorphism by simple (TTTA) repetition of the gene CYP19 in healthy women and women with mammary carcinoma. Material and methods: We conducted a study among 180 healthy women and 70 women with breast cancer.The DNA of the leukocytes was obtained for the genotyping TTTA’s repetitions of CYP19.They were determined by amplification by PCR, the amplified products moved along in gel of acrilamide. Results: 250 women (age 55 ± 12 years) went included. BMI 30 ± 7.1 Kg/m.2 Found the distribution of the different frequencies of CYP19 allele, in healthy women the allele with 6 (32.7 %) and 7 (21.6 %) repetitions were the most frequent, in women with breast cancer the allele with 6(29%) and 10(26%)repetitions were the most frequent.
