Resumen en español El objetivo de esta investigación fue evaluar dos grupos de oligonucleótidos para la detección de dos mutaciones puntuales en el gen de la acetil–CoA carboxilasa (ACCasa) de Avena fatua L., durante el período 2005–2006, las cuales están relacionadas con la resistencia a herbicidas que inhiben la síntesis de ácidos grasos. Se seleccionaron cuatro poblaciones de avena silvestre (M5r y M28r) resistentes a la aplicación en campo del herbicida clodinafop propargil y dos poblaciones susceptibles (M25s y M43s). Se desarrollaron dos grupos de oligonucleótidos con base en la secuencia del gen de la ACCasa de la avena silvestre (Gene Bank No. de acceso AF231334). El primer grupo de oligonucleótidos, VSE11GM, VRE11RGM, ACVII11GM y ACVII11RGM, se diseñaron para detectar el cambio de timina (T) por adenina (A) en la posición 1 322 del gen descrito y originando el cambio de isoleucina (ATT) por asparagina (AAT). El segundo grupo de oligonucleótidos, VRDICGM, VRDITRGM, ACVRG1GM y ACVRG1RGM, se diseñaron para detectar el cambio de timina (T) o citosina (C) por adenina (A) en la posición 541 del mismo gen y originando el cambio de isoleucina (ATA) por leucina (CTA o TTA). No se logró diferenciar las poblaciones que presentaron resistencia en campo de las que fueron susceptibles, lo que causaría el cambio de isoleucina por leucina en posición 541 del gen para ambas mutaciones. La ausencia de la amplificación del fragmento de 480 pb para la población resistente M5r, con el primer grupo de oligonucleótidos, así como la ausencia de la amplificación de los fragmentos de 496 pb y 330 pb en las cuatro poblaciones con el segundo grupo de oligonucleótidos, indica la probabilidad de que sea una sustitución diferente la que le confiere resistencia a la avena silvestre hacia este grupo de herbicidas
Resumen en inglés Two oligonucleotid groups were evaluated in order to detect two punctual mutations of the gene acetyl–CoA carboxilase (ACCasa) in Avena fatua L., which are related to the resistance to herbicides that inhibit fatty acid synthesis. Four populations of wild oat, two resistant (M5r and M28r) and two susceptible (M25s and M43s), to field application of clodinafop propargil were selected. Two groups of oligonucleotids were developed based on the sequence of the gene of the ACCasa from wild oat (GeneBank access number AF231334). The first group, VSE11GM, VRE11RGM, ACVII 11GM and ACVII 11RGM, was designed to detect the change of timine (T) by adenine (A) in position 1 322 of the described gene and which originated the change of isoleucine (ATT) by asparagine (AAT). The second group of oligonucleotids, VRDICGM, VRDITRGM, ACVRG1GM and ACVRG1RGM, was designed to detect the change of citosine or timine (T or C) by adenine (A) in position 541 of the same gene, and which originated the change of isoleucine (ATA) by leucine (CTA or TTA). Consequently, it was not possible to differentiate the populations that offered resistance in field from those which were susceptible and which would cause the change of isoleucine by leucine in the position 541 of the gene for both mutations. The absence of amplification of the fragment of 480 pb for the resistant population M5r, with the first group of oligonucleotids, as well as the absence of the amplification of the fragments 496 pb and 330 pb in the four populations with the second group of oligonucleotids, indicated the probability that it is a different substitution the one that confers resistance to the wild oat toward this group of herbicides
